Cisplatin + everolimus is a combination therapy regimen investigated for the treatment of advanced extrapulmonary neuroendocrine carcinoma (EP-NEC). This regimen combines cisplatin, a platinum-based cytotoxic chemotherapy that induces DNA cross-linking and subsequent apoptosis, with everolimus, a targeted inhibitor of the mammalian target of rapamycin (mTOR) pathway. The mTOR pathway is frequently dysregulated in neuroendocrine tumors, playing a critical role in cell growth, metabolism, and angiogenesis. The combination was evaluated in a Phase II clinical trial (NCT02695459) sponsored by the Netherlands Cancer Institute, where patients received intravenous cisplatin every three weeks and daily oral everolimus to assess antitumor activity and safety as a first-line treatment.
