Overview

Cisplatin + fluorouracil + radiotherapy is a combination therapy used primarily in the treatment of various locally advanced or recurrent cancers such as esophageal squamous cell carcinoma, head and neck cancer, and bladder cancer. Cisplatin is a platinum-based chemotherapeutic agent that acts mainly by forming DNA crosslinks and inhibiting DNA synthesis and function. Fluorouracil (also known as 5-FU) is an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis. When combined with radiotherapy (the use of ionizing radiation to kill tumor cells), both drugs act as radiosensitizers—making cancer cells more susceptible to the effects of radiation by interfering with DNA repair mechanisms and cell cycle checkpoints. This trimodality approach has demonstrated improved response rates compared to single-modality treatments in several cancers[1][2][3][4][5][6].
