Overview

This therapeutic regimen is designed for the treatment of advanced nasopharyngeal carcinoma (NPC), specifically targeting patients with N2-3 stage disease. The protocol consists of concurrent chemoradiotherapy (CCRT) followed by adjuvant chemotherapy. During the CCRT phase, patients receive concurrent cisplatin (a platinum-based DNA cross-linking agent) alongside intensity-modulated radiotherapy (IMRT). This is followed by an adjuvant chemotherapy phase known as the PF regimen, which combines cisplatin with 5-fluorouracil (5-FU). 5-fluorouracil is a pyrimidine analog antimetabolite that inhibits thymidylate synthase, thereby interfering with DNA synthesis. This specific treatment sequence was evaluated in a Phase 3 clinical trial conducted by Sun Yat-sen University to compare its efficacy and safety against a gemcitabine plus cisplatin (GP) adjuvant regimen.
