This drug is a combination chemotherapy regimen consisting of three agents: - **Cisplatin** is a platinum-based chemotherapeutic agent that causes DNA crosslinking and damage, leading to apoptosis in rapidly dividing cancer cells. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase, resulting in cell death. - **Gefitinib** is an orally active small molecule epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that blocks EGFR signaling pathways involved in tumor cell proliferation and survival. This combination has been studied primarily for advanced or metastatic non-small-cell lung cancer (NSCLC) and advanced urothelial carcinoma. Clinical trials have shown the combination to be generally well tolerated but did not demonstrate improved efficacy over gemcitabine plus cisplatin alone in NSCLC patients[1][2]. Gefitinib targets the EGFR pathway, which is often overexpressed or mutated in certain cancers, aiming to enhance chemotherapy effects through targeted therapy.
