Overview

This is a combination regimen comprising three agents: **cisplatin**, **gemcitabine**, and **interferon-alpha (IFN-alpha)**. Cisplatin is a platinum-based chemotherapeutic that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. Gemcitabine is a nucleoside analog that is incorporated into DNA, inhibiting DNA synthesis and triggering cell death. Interferon-alpha is a cytokine that modulates immune responses, has direct anti-proliferative effects, and can enhance the antitumor activity of chemotherapy. This combination has been investigated, particularly in the setting of advanced or metastatic solid tumors, such as pancreatic and neuroendocrine cancers, in regimens sometimes including adjunctive thermal therapy. The regimen is typically administered intravenously, with cisplatin and gemcitabine given in a schedule-optimized sequence and interferon-alpha given daily at low doses. Early-phase trials demonstrated partial and complete responses, especially in cancers resistant to standard treatment, with notable toxicities including peripheral neuropathy and ototoxicity[1][4].
