This is a combination therapy consisting of three agents: - **Cisplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks and inhibits DNA synthesis and function, leading to apoptosis in rapidly dividing cells. - **Gemcitabine** is a nucleoside analog (antimetabolite) that incorporates into DNA during replication, causing chain termination and cell death. - **Kanglaite** is an injectable emulsion derived from Coix seed oil (traditional Chinese medicine), containing triglycerides with four types of fatty acids. It exhibits antitumor activity by inducing apoptosis via the PI3K/AKT pathway, enhancing immune responses (increasing CD3+, CD4+, and CD4+/CD8+ ratios), reversing multidrug resistance, improving patient performance status (KPS score), and reducing chemotherapy-related adverse effects. The combination has been primarily studied as first-line or adjunctive therapy for advanced non-small cell lung cancer (NSCLC). Meta-analyses show it increases objective response rate, disease control rate, immune function markers, quality of life scores (KPS), and reduces adverse reactions compared to chemotherapy alone[1][2][3][5]. Kanglaite has also been approved in China for various malignancies including NSCLC; it was the first traditional Chinese medicine preparation approved by the US FDA for clinical trials in solid tumors[7][10].
