This is a combination therapy consisting of cisplatin, a platinum-based antineoplastic agent, and monosialoganglioside (GM1), a neuroprotective glycosphingolipid. Cisplatin acts as an alkylating agent that cross-links DNA strands, inhibiting DNA replication and transcription, leading to apoptosis in rapidly dividing cancer cells. It is widely used in the treatment of various cancers but is associated with significant neurotoxicity among other side effects[3][5]. Monosialoganglioside (GM1) has been investigated for its potential to prevent or reduce chemotherapy-induced peripheral neuropathy (CIPN), particularly the neurotoxic effects caused by agents like cisplatin. GM1 may exert its protective effect by modulating neuronal repair mechanisms and reducing oxidative stress in neural tissues[6][7]. The combination has been studied primarily for its ability to maintain anticancer efficacy while mitigating the dose-limiting neurotoxicity of cisplatin-based chemotherapy regimens.
