This is a four-drug chemotherapy combination consisting of cisplatin, nab-paclitaxel (nanoparticle albumin-bound paclitaxel), capecitabine, and gemcitabine. Each component has a distinct mechanism of action targeting cancer cell proliferation and survival: - **Cisplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks, leading to apoptosis. - **Nab-paclitaxel** is an albumin-bound formulation of paclitaxel that stabilizes microtubules and inhibits their disassembly, disrupting mitosis. - **Capecitabine** is an oral prodrug converted to 5-fluorouracil (5-FU) in the body; it inhibits thymidylate synthase, impairing DNA synthesis. - **Gemcitabine** is a nucleoside analog that incorporates into DNA during replication and inhibits ribonucleotide reductase. This combination brings together agents with complementary mechanisms for enhanced antitumor activity. It has been studied primarily in advanced or metastatic solid tumors such as pancreatic cancer, biliary tract cancers, and nasopharyngeal carcinoma[2][4][6]. The regimen aims to maximize cytotoxic effects while managing overlapping toxicities.
