This is a combination regimen consisting of three agents: - **Cisplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. - **Pemetrexed** is an antifolate antineoplastic agent that inhibits multiple folate-dependent enzymes involved in the synthesis of thymidine and purine nucleotides, thereby disrupting DNA and RNA synthesis essential for cell replication[1][4][5]. - **Panitumumab** is a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand-induced activation and downstream signaling pathways involved in cell proliferation and survival. This combination has been explored primarily for advanced non-squamous non-small cell lung cancer (NSCLC), particularly in patients with KRAS wild-type tumors. The addition of panitumumab to standard chemotherapy regimens such as pemetrexed plus platinum compounds has been investigated to improve outcomes by combining cytotoxic effects with targeted EGFR inhibition. However, clinical studies indicate increased toxicity without clear improvement in efficacy when panitumumab is added[6].
