Overview
Cisplatin + pemetrexed + bevacizumab is a combination chemotherapy regimen used primarily as first-line treatment for advanced or metastatic non-squamous non-small cell lung cancer (NSCLC) and unresectable malignant pleural mesothelioma. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis. Pemetrexed is an antifolate that inhibits enzymes involved in purine and pyrimidine synthesis, disrupting DNA replication and cell division. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor (VEGF), thereby inhibiting angiogenesis required for tumor growth. This combination has demonstrated favorable efficacy and manageable toxicity profiles in clinical studies, with maintenance therapy using pemetrexed plus bevacizumab often following induction cycles[1][2][5][6].
