This is a combination therapy consisting of cisplatin, pemetrexed, and recombinant human endostatin (rh-endostatin). Cisplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. Pemetrexed is an antifolate antineoplastic agent that inhibits multiple folate-dependent enzymes involved in nucleotide synthesis, thereby blocking DNA replication and cell division. Rh-endostatin (commonly known by the brand name Endostar) is a recombinant form of endostatin, an endogenous inhibitor of angiogenesis; it works by inhibiting the proliferation and migration of endothelial cells required for new blood vessel formation within tumors. This combination has been primarily investigated as first-line or maintenance therapy for advanced non-small cell lung cancer (NSCLC), with studies suggesting potential benefits in progression-free survival when used as maintenance but not consistently showing significant improvement over chemotherapy alone in all settings[1][3][5]. The regimen leverages cytotoxic effects from cisplatin/pemetrexed alongside anti-angiogenic activity from rh-endostatin.
