Overview

Cisplatin + CKM + celecoxib + DC vaccine is an investigational chemo-immunotherapy combination regimen developed by the Roswell Park Comprehensive Cancer Center for the treatment of advanced-stage ovarian cancer. The regimen is designed to reprogram the tumor microenvironment (TME) from an immunosuppressive state to one that favors the infiltration and activity of cytotoxic T lymphocytes (CTLs). It consists of four primary components: intraperitoneal cisplatin (a DNA-alkylating chemotherapy), a chemokine modulatory (CKM) regimen comprising rintatolimod (a TLR3 agonist) and interferon-alpha (IFNα), oral celecoxib (a COX-2 inhibitor), and an autologous alpha-type-1-polarized dendritic cell (alpha-DC1) vaccine. By combining these agents, the therapy aims to increase the production of CTL-attracting chemokines (such as CXCL9 and CXCL10) while suppressing regulatory T cell (Treg)-attracting signals (such as CCL22) and prostaglandin E2 (PGE2) production. This multi-pronged approach is currently being evaluated in Phase 1/2 clinical trials for patients with recurrent or advanced ovarian cancer.
