Overview

Cisplatin + sodium thiosulfate is an investigational combination regimen developed by the University of California, San Diego (UCSD) for the treatment of primary lung cancer and lung metastases. The protocol involves the supradose intra-arterial (IA) administration of cisplatin directly into the bronchial or pulmonary arteries supplying the tumor, paired with the simultaneous intravenous (IV) administration of sodium thiosulfate. Cisplatin is a platinum-based chemotherapeutic agent that induces apoptosis by forming DNA crosslinks. Sodium thiosulfate acts as a systemic neutralizing or "rescue" agent, binding to and inactivating cisplatin in the systemic circulation to mitigate nephrotoxicity and other systemic side effects while maintaining high local concentrations at the tumor site. This approach was evaluated in a Phase I/Pilot study (NCT01114958) to characterize toxicity and radiographic response.
