Overview
Cisplatin + temozolomide is a combination chemotherapy regimen consisting of two small molecule alkylating agents. Cisplatin is a platinum-based compound that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. Temozolomide is an imidazotetrazine derivative that methylates DNA at the O6 and N7 positions of guanine, resulting in cytotoxicity and cell death. This combination has been investigated primarily for use in metastatic melanoma and recurrent glioblastoma multiforme, with the rationale that cisplatin may enhance the activity of temozolomide by inhibiting DNA repair mechanisms such as MGMT. The regimen has shown modest efficacy but increased toxicity compared to single-agent therapy[1][3].
