Cisplatin + veliparib is a combination therapy consisting of cisplatin, a platinum-based chemotherapeutic agent, and veliparib, a potent inhibitor of Poly (ADP-ribose) polymerase (PARP). This combination works through complementary mechanisms - cisplatin causes DNA damage by forming crosslinks in DNA, while veliparib inhibits PARP enzymes that are involved in DNA repair. By preventing cancer cells from repairing DNA damage caused by cisplatin, veliparib potentiates the cytotoxic effects of chemotherapy. This synergistic effect is particularly pronounced in cancers with deficiencies in homologous recombination repair, such as those with BRCA mutations.
