Cisplatin + vinflunine is a chemotherapy combination regimen that pairs a platinum-based agent (cisplatin) with a third-generation bifluorinated semi-synthetic vinca alkaloid (vinflunine). Vinflunine works primarily by binding to tubulin and disrupting microtubule dynamics, leading to mitotic arrest and cell death. This combination has shown promising activity in advanced non-small cell lung cancer (NSCLC) and has been investigated for use in advanced urothelial carcinoma (bladder cancer). In a phase I/II study in NSCLC, the recommended dose was established at cisplatin 80 mg/m² combined with vinflunine 320 mg/m², with a tumor response rate of 32.1% and disease control achieved in 79.2% of patients. The combination has also been proposed as a potential neoadjuvant treatment option for bladder cancer patients.
