Citalopram + trazodone is a combination of two antidepressant medications, each with distinct mechanisms of action. Citalopram is a selective serotonin reuptake inhibitor (SSRI) that increases serotonin levels in the brain by inhibiting its reuptake into presynaptic cells, thereby enhancing serotonergic neurotransmission. Trazodone is classified as a serotonin antagonist and reuptake inhibitor (SARI); it acts primarily by antagonizing serotonin 5-HT2A receptors and weakly inhibiting the reuptake of serotonin, also increasing its availability in the synaptic cleft. This combination may be used off-label for co-occurring depression and insomnia but carries an increased risk for adverse effects such as serotonin syndrome due to additive serotonergic activity[5][6].
