A combination therapy consisting of cixutumumab, a fully human immunoglobulin G1 monoclonal antibody directed against the type 1 insulin-like growth factor receptor (IGF-1R), and temsirolimus, a mammalian target of rapamycin (mTOR) inhibitor. This combination was developed based on the rationale that mTOR inhibitors mediate AKT activation through an IGF-1R-dependent mechanism, and combining these agents could enhance anticancer activity by modulating resistance to mTOR inhibition.
