Overview

CJ-040,714 (also known as PF-0040714) is a small molecule potassium-competitive acid blocker (P-CAB) that was developed by Pfizer in collaboration with CJ Healthcare (now HK inno.N) for the treatment of gastro-esophageal reflux disease (GERD). As a P-CAB, it functions by reversibly inhibiting the gastric H+/K+-ATPase (the proton pump) in a potassium-competitive manner, offering a more rapid onset of action and more stable acid suppression compared to traditional proton pump inhibitors (PPIs). The compound was evaluated in Phase 2 clinical trials to assess its efficacy in reducing distal esophageal acid exposure and the duration of reflux events in patients with erosive GERD. However, development of the asset was discontinued following these early-stage clinical evaluations.
