Clade B gag DNA + env DNA is an investigational prophylactic HIV-1 vaccine candidate composed of two separate DNA plasmids encoding the Gag and Env proteins from HIV-1 Clade B. The vaccine is designed to induce both cellular (T-cell) and humoral (antibody) immune responses against HIV-1 by delivering genetic material that expresses these viral antigens in host cells. This approach aims to stimulate the immune system to recognize and respond to key structural proteins of the virus, potentially providing protection against infection or modulating disease progression. The combination has been studied as a prime in prime/boost regimens, sometimes followed by protein or viral vector boosts[3][4][6]. It has shown immunogenicity in early-phase clinical trials, eliciting both T-cell responses (notably IFN-gamma producing CD4+ and CD8+ T-cells) and binding antibodies targeting multiple regions of Env as well as Gag[6]. Development has primarily been led by public sector organizations such as the National Institute of Allergy & Infectious Diseases.
