Clade C gp140 + adjuvant is a combination vaccine candidate composed of a trimeric, recombinant envelope glycoprotein 140 (gp140) derived from HIV-1 subtype (clade) C and an immunological adjuvant—most commonly aluminum phosphate or other clinically relevant adjuvants such as MF59 or GLA-SE. The vaccine is designed to elicit immune responses against the envelope protein of HIV-1 subtype C, which is the most prevalent strain in regions such as sub-Saharan Africa. The mechanism of action involves stimulating both humoral and cellular immunity by presenting the stabilized trimeric form of the viral envelope to the immune system in conjunction with an adjuvant that enhances immunogenicity and antibody response breadth[1][3][6]. This formulation has been evaluated alone or as part of heterologous prime/boost regimens with viral vectors (e.g., Ad26.Mos4.HIV), often in combination with mosaic antigens to broaden cross-clade protection[4][7][9]. Clinical trials have shown that adding mosaic proteins to this regimen increases immune response breadth without compromising safety[1].
