Overview

Cladribine + cytarabine is a chemotherapeutic combination used primarily for the treatment of acute myeloid leukemia (AML). Cladribine is a purine analogue that exerts cytotoxic, proapoptotic, and antiproliferative effects on AML cells and inhibits DNA repair. Cytarabine is a pyrimidine nucleoside analogue that inhibits DNA synthesis. The combination works synergistically, as cladribine increases cytarabine triphosphate levels in leukemic cells, enhancing cytarabine's effectiveness. This combination has been studied in various clinical settings including newly diagnosed AML, elderly patients unfit for intensive chemotherapy, and relapsed/refractory AML.
