Overview

This is a combination chemotherapy regimen consisting of cladribine (a purine nucleoside analog), cytarabine (an antimetabolite), and granulocyte colony stimulating factor (G-CSF, a hematopoietic growth factor). The combination is primarily used in the treatment of relapsed or refractory acute myeloid leukemia (R/R AML). Cladribine inhibits DNA synthesis by incorporating into DNA and causing strand breaks. Cytarabine also inhibits DNA synthesis by acting as an antimetabolite. G-CSF stimulates the production of neutrophils and may sensitize leukemic cells to chemotherapy while supporting recovery from myelosuppression. This regimen has demonstrated significant efficacy in R/R AML with high remission rates and manageable toxicity profiles[1][5][6].
