Overview

A combination chemotherapy regimen consisting of cladribine, homoharringtonine, and cytarabine. This regimen has shown promising results in treating acute myeloid leukemia (AML), particularly in patients with de novo AML. The combination works through synergistic mechanisms: cladribine can increase cytarabine triphosphate levels, enhancing its effectiveness[6]. Homoharringtonine has been shown to target leukemia diseases that have switched their anti-apoptotic dependence from BCL-2 to MCL-1[1]. Together, these agents demonstrate synergistic antiproliferative effects against AML cells[2]. Clinical trials have demonstrated high remission rates, including in patients with adverse risk factors such as specific cytogenetic abnormalities.
