Clascoterone + adapalene is a topical combination therapy used off-label for the treatment of acne vulgaris. Clascoterone is a first-in-class topical androgen receptor inhibitor that works by competing with dihydrotestosterone (DHT) for binding to androgen receptors in sebaceous glands and hair follicles, thereby reducing sebum production and inflammation associated with acne[6][8]. Adapalene is a synthetic retinoid-like compound that modulates cellular differentiation, keratinization, and inflammatory processes in the skin[3]. The combination targets multiple pathogenic factors in acne—sebum production, follicular hyperkeratinization, inflammation—offering an additive or synergistic effect.
