Claudin18.2-targeted CAR-T cells are an investigational autologous cell therapy developed by the Second Affiliated Hospital, School of Medicine, Zhejiang University. The therapy consists of T cells genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets Claudin 18.2 (CLDN18.2), a tight junction protein that is highly expressed in several gastrointestinal cancers, including colorectal cancer, but has limited expression in normal tissues. In clinical development, these CAR-T cells are being evaluated in combination with Claudin18.2-targeted CAR-modified dendritic cells (CAR-DCs). This combination approach aims to improve the therapeutic efficacy of the CAR-T cells by using the CAR-DCs to stimulate T-cell activation and persistence within the immunosuppressive environment of advanced solid tumors.
