CLD-101 (also known as NSC-CRAd-S-pk7) is a first-in-class oncolytic virotherapy that utilizes a neural stem cell (NSC) delivery system to treat high-grade gliomas. Developed by City of Hope and licensed to Calidi Biotherapeutics, the therapy consists of the HB1.F3.CD21 immortalized NSC line loaded with CRAd-S-pk7, a conditionally replicating adenovirus. The NSCs act as Trojan horses, migrating through the brain parenchyma to reach invasive tumor cells and protecting the viral payload from the host immune system. The CRAd-S-pk7 virus is engineered with a survivin promoter to restrict replication to malignant cells and a polylysine-modified fiber (pk7) to enhance viral entry. Upon reaching the tumor, the virus replicates, induces cell lysis, and triggers a secondary anti-tumor immune response.
