Overview

CLDN18.2 + CD19 CAR-T is an investigational autologous chimeric antigen receptor T cell (CAR-T) therapy designed to target both Claudin 18.2 (CLDN18.2), a tight junction protein highly expressed in certain gastrointestinal cancers, and CD19, a B-cell marker commonly targeted in hematologic malignancies[4]. This dual-targeting approach aims to enhance anti-tumor efficacy by addressing tumor heterogeneity and potentially overcoming resistance mechanisms seen with single-antigen targeting therapies. Preclinical data published by BioNTech demonstrated the feasibility of generating CAR-T cells that simultaneously target both CLDN18.2 and CD19 antigens, showing promising activity against solid tumors expressing these targets[4]. The therapy is being developed primarily for the treatment of advanced solid tumors such as gastric cancer and pancreatic cancer, as well as for hematologic malignancies where CD19 expression is relevant.
