Overview

Clevudine + adefovir dipivoxil is an investigational combination of two antiviral agents used for the treatment of chronic hepatitis B virus (HBV) infection. Clevudine is a nucleoside analog that inhibits HBV DNA polymerase, thereby suppressing viral replication. Adefovir dipivoxil is a diester prodrug of adefovir, an acyclic nucleotide analog of adenosine monophosphate. After conversion to its active metabolite, adefovir diphosphate, it inhibits HBV DNA polymerase (reverse transcriptase) by competing with deoxyadenosine triphosphate and causing chain termination after incorporation into viral DNA[2][3][5]. The combination aims to enhance antiviral efficacy and reduce the risk of resistance in patients with chronic hepatitis B.
