Overview

Clindamycin + dapsone is a combination of two small molecule antibiotics used primarily in dermatology, most notably for the treatment of acne vulgaris. Clindamycin is a lincosamide antibiotic that inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, providing activity against susceptible anaerobic bacteria and some Gram-positive organisms[2]. Dapsone is a sulfone antibiotic with both antibacterial and anti-inflammatory properties; its mechanism in acne is not fully understood but includes inhibition of neutrophil myeloperoxidase and reduction of inflammatory mediators[6][7]. Both agents are available as topical formulations for acne, though they are more commonly combined with other agents (such as benzoyl peroxide or retinoids) rather than together. The rationale for combining clindamycin and dapsone would be to leverage their complementary antibacterial and anti-inflammatory effects to improve outcomes in patients with moderate-to-severe or treatment-resistant acne.
