**Clivatuzumab tetraxetan + gemcitabine** is an investigational drug combination used primarily in the treatment of metastatic pancreatic cancer. It comprises **clivatuzumab tetraxetan**—a humanized monoclonal antibody-drug conjugate targeting mucins (primarily MUC1/MUC5AC) expressed in pancreatic carcinoma—with tetraxetan as a chelator for the therapeutic radioisotope yttrium-90 (^90Y)[4][6]. The antibody delivers ^90Y directly to tumor cells, resulting in localized radiotherapeutic destruction. The combination includes **gemcitabine**, a nucleoside analog antimetabolite that acts as a radiosensitizer, enhancing the cytotoxic effect of the radioimmunotherapy and contributing direct antineoplastic activity. This regimen is designed for patients who have progressed following standard therapies. Early clinical studies showed modest response rates and manageable myelosuppression, but did not ultimately demonstrate improved overall survival in a phase III trial, which was terminated early[4][3][7]. The combination was developed by Immunomedics.
