CLL1:TRAIL (also known as scFvCLL1:TRAIL) is a recombinant fusion protein biologic designed to enhance the tumoricidal activity of granulocytes. It consists of a single-chain variable fragment (scFv) antibody targeting the myeloid marker C-type lectin-like molecule-1 (CLL1/CLEC12A) fused to the extracellular domain of the pro-apoptotic protein tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). The mechanism involves selectively binding to CLL1 on the surface of granulocytes and monocytes, thereby "arming" these immune effector cells with high levels of membrane-associated TRAIL. This allows the leukocytes to trigger apoptosis in cancer cells via TRAIL receptors (DR4 and DR5) and significantly potentiates the antibody-dependent cell-mediated cytotoxicity (ADCC) of therapeutic monoclonal antibodies such as rituximab and cetuximab. It is primarily investigated as an adjuvant to optimize anticancer antibody therapies.
