CLL1 CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy targeting C-type lectin-like molecule-1 (CLL1, also known as CLEC12A), a surface antigen highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells, but absent on normal hematopoietic stem cells. Developed by researchers led by Donghua Zhang at Tongji Hospital (Huazhong University of Science and Technology), this therapy is specifically designed for patients with relapsed or refractory AML. In clinical studies, it is often utilized as a sequential treatment strategy where the CAR-T infusion is followed by allogeneic hematopoietic stem cell transplantation (allo-HSCT). This approach aims to achieve deep molecular remission, effectively bridging the patient to a curative transplant while minimizing the risk of post-transplant relapse by eradicating the leukemic stem cell reservoir.
