Overview

CLL1 CAR-T + CLL1+CD33 CAR-T is a combination of two investigational chimeric antigen receptor T cell (CAR-T) therapies designed for the treatment of acute myeloid leukemia (AML). The first component, CLL1 CAR-T, consists of autologous or allogeneic T cells engineered to express a chimeric antigen receptor targeting the cell surface protein C-type lectin-like molecule-1 (CLL-1/CLEC12A), which is highly expressed on AML blasts and leukemic stem cells but not on normal hematopoietic stem cells. The second component, CLL1+CD33 CAR-T, refers to dual-targeting or co-administered T cells engineered to recognize both CLL-1 and CD33 antigens—another myeloid marker frequently found in AML. By simultaneously targeting these two antigens, this combination aims to enhance anti-leukemic efficacy and reduce the risk of disease relapse due to antigen escape. Both modalities function by redirecting patient or donor-derived T lymphocytes toward malignant AML cells expressing these targets, leading to their activation and cytotoxic destruction through immune-mediated mechanisms[6][8][9]. These therapies are under early-phase clinical investigation for relapsed/refractory AML.
