CLN-619 + datopotamab deruxtecan

A combination of **CLN-619**, a humanized IgG1 monoclonal antibody targeting MICA and MICB, and **datopotamab deruxtecan**, a Trop-2-directed antibody-drug conjugate (ADC) with a DNA topoisomerase I inhibitor payload, under investigation for oncology indications. **CLN-619** binds to the alpha-3 domain of MICA/B, prevents ligand shedding, increases cell surface expression, and induces antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis (ADCP) by engaging NKG2D and CD16A receptors on NK cells and macrophages. **Datopotamab deruxtecan** targets Trop-2, is internalized, and releases its cytotoxic topoisomerase I inhibitor payload to induce DNA breaks and apoptosis in Trop-2 expressing tumor cells.
