Clobetasol propionate + calcitriol is a combination treatment regimen developed by Galderma for the management of moderate plaque psoriasis. The regimen utilizes two distinct topical agents: Clobex (clobetasol propionate 0.05% spray), a super-high-potency corticosteroid, and Vectical (calcitriol 3 mcg/g ointment), a vitamin D3 analog. Clobetasol propionate provides rapid anti-inflammatory action by activating glucocorticoid receptors, which suppresses the production of inflammatory cytokines. Calcitriol acts as an agonist at the vitamin D receptor (VDR), inhibiting the hyperproliferation of keratinocytes and inducing their differentiation. This dual approach is designed to control acute flares with the corticosteroid while providing long-term maintenance and minimizing steroid-related side effects through the use of the vitamin D analog.
