Overview

A combination chemotherapy regimen consisting of clofarabine (a purine nucleoside antimetabolite), cyclophosphamide (an alkylating agent), and alemtuzumab (a monoclonal antibody targeting CD52). This combination is used in the treatment of relapsed or refractory leukemia, particularly in cases where standard therapies have failed. Each component has a distinct mechanism of action: clofarabine interferes with DNA synthesis and repair, cyclophosphamide damages DNA by forming cross-links, and alemtuzumab targets CD52-expressing lymphocytes causing cell death through antibody-dependent cellular cytolysis and complement-mediated lysis.
