Overview

CLARA is a combination chemotherapy regimen developed by Hospices Civils de Lyon for the treatment of acute myeloid leukemia (AML). The regimen consists of clofarabine, a second-generation purine nucleoside analog, and intermediate-dose cytarabine (ara-C), a pyrimidine nucleoside analog, typically administered alongside G-CSF (granulocyte colony-stimulating factor). Clofarabine acts by inhibiting DNA polymerase and ribonucleotide reductase, while cytarabine is incorporated into DNA, leading to the inhibition of DNA polymerase and termination of DNA strand elongation. This combination is specifically being evaluated in a randomized Phase II trial as a consolidation therapy for younger patients (18-60 years) with newly diagnosed intermediate or unfavorable risk AML, comparing its efficacy and toxicity against standard high-dose cytarabine (HDAC).
