This combination therapy consists of clomiphene citrate and dexamethasone and is being evaluated for ovulation induction in women with infertility undergoing intrauterine insemination (IUI). Clomiphene citrate is a selective estrogen receptor modulator (SERM) that induces ovulation by inhibiting negative feedback of estrogen on the hypothalamus, thereby increasing the secretion of gonadotropins. Dexamethasone, a potent corticosteroid and glucocorticoid receptor agonist, is added to the regimen to enhance clomiphene's efficacy, particularly in clomiphene-resistant patients, by suppressing adrenal androgen production. This specific investigational combination was studied in a Phase 4 clinical trial (NCT01856062) led by the San Antonio Military Medical Center at Brooke Army Medical Center.
