Overview

Clopidogrel + everolimus is a combination of two small molecule drugs used primarily in research and clinical settings to prevent transplant arteriosclerosis and for dual antiplatelet therapy after stent implantation. Clopidogrel is an oral antiplatelet agent that acts as an antagonist of the P2Y12 adenosine diphosphate receptor on platelets, thereby inhibiting platelet aggregation. Everolimus is an inhibitor of the mammalian target of rapamycin (mTOR), which suppresses cell proliferation and immune responses. The combination has been shown in preclinical models to dramatically reduce neointimal hyperplasia and transplant arteriosclerosis, likely due to synergistic inhibition of platelet activation and vascular smooth muscle cell proliferation[1][2][4]. In clinical practice, this combination may be encountered when clopidogrel is used as part of dual antiplatelet therapy following percutaneous coronary intervention with everolimus-eluting stents[6].
