CM24 + nivolumab

CM24 + nivolumab is a combination immunotherapy regimen under investigation for the treatment of advanced solid tumors, particularly metastatic pancreatic ductal adenocarcinoma (PDAC). CM24 is a monoclonal antibody that inhibits Carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1), an immune checkpoint protein implicated in tumor immune evasion. Nivolumab is a fully human IgG4 monoclonal antibody targeting Programmed cell death protein 1 (PD-1), another key immune checkpoint. By blocking both CEACAM1 and PD-1 pathways, this combination aims to enhance anti-tumor T-cell activity and overcome resistance to single-agent immunotherapies. The regimen has shown encouraging safety and efficacy signals in early-phase clinical trials, including improved overall survival compared to historical controls in heavily pretreated PDAC patients[3][4][5]. The combination is being developed by Purple Biotech (for CM24) and Bristol Myers Squibb (for nivolumab).
