Overview

CMB305 + atezolizumab is an investigational **combination therapy** for the treatment of advanced or metastatic synovial sarcoma or myxoid/round cell liposarcoma expressing the NY-ESO-1 antigen. **CMB305** is a heterologous prime-boost cancer vaccine regimen composed of LV305 (a dendritic cell–targeted, non-integrating lentiviral vector encoding NY-ESO-1) and G305 (a recombinant NY-ESO-1 protein formulated with glucopyranosyl lipid A, a TLR4 agonist adjuvant). **Atezolizumab** is a monoclonal antibody targeting PD-L1 (programmed death-ligand 1), acting as an immune checkpoint inhibitor. The rationale for combining these agents is to induce robust NY-ESO-1–specific T-cell and antibody responses via vaccination, while blocking PD-L1 to prevent tumor-mediated immune evasion, thereby enhancing anti-tumor immunity. The combination aims to improve progression-free and overall survival compared to immune checkpoint inhibition alone, though pivotal trials did not show statistically significant survival benefit. Immune response biomarkers suggest enhanced anti-NY-ESO-1 T-cell and antibody induction with combination therapy over atezolizumab alone[1][5][7].
