Drug intelligence / Profile preview
CN54gp140 + GLA-AF is a recombinant protein-based HIV-1 vaccine candidate developed by Imperial College London. It consists of a trimeric envelope glycoprotein derived from a clade C HIV-1 strain (CN54), which is the most prevalent subtype in the global epidemic. The vaccine is formulated with GLA-AF (glucopyranosyl lipid A-aqueous formulation), a synthetic Toll-like receptor 4 (TLR4) agonist adjuvant designed to enhance the magnitude and breadth of the immune response. The vaccine aims to elicit both systemic and mucosal antibodies to prevent HIV-1 infection. Clinical evaluation has focused on Phase 1 trials assessing safety, tolerability, and immunogenicity in healthy, HIV-uninfected adults.
