Drug intelligence / Profile preview
CN54gp140 + GLA-AF is an experimental HIV-1 vaccine candidate consisting of a recombinant trimeric envelope glycoprotein (gp140) and a synthetic adjuvant. The immunogen, CN54gp140, is derived from the HIV-1 clade C strain 97CN54, which represents a predominant subtype in regions heavily affected by the HIV epidemic. The vaccine is formulated with GLA-AF (glucopyranosyl lipid A-aqueous formulation), a synthetic Toll-like receptor 4 (TLR4) agonist that enhances the magnitude and quality of the immune response by promoting innate immune activation. Developed by researchers at Imperial College London, this vaccine candidate is designed to elicit both systemic and mucosal humoral responses, as well as cellular immunity, for the prevention of HIV infection in healthy individuals. It has been evaluated in Phase 1 clinical trials to assess its safety, tolerability, and immunogenicity using various intramuscular prime-boost regimens.
