CN54gp140 + hsp70 is an experimental combination vaccine designed to prevent infection with Human Immunodeficiency Virus (HIV). The formulation consists of a synthetically produced HIV-1 envelope glycoprotein (CN54gp140) covalently linked to the human heat shock protein 70 (hsp70). The gp140 component serves as the antigen, while hsp70 acts as an immunological adjuvant, enhancing immune responses. This conjugate is intended for mucosal administration, specifically via cervico-vaginal topical application in women, aiming to induce local and systemic immunity at the primary site of HIV entry. Preclinical studies and a Phase I clinical trial have evaluated its safety and immunogenicity in healthy female volunteers[1][2]. Mechanistically, the vaccine stimulates both humoral (IgG/IgA) and cellular immune responses against HIV-1 antigens; hsp70 also activates innate immunity by upregulating CC chemokines and modulating CCR5 expression on CD4+ T cells[1].
