CN54gp140 + MPLA is a combination vaccine candidate designed for the prevention of HIV infection. It consists of two components: recombinant CN54gp140, which is an HIV-1 envelope glycoprotein derived from the clade C strain 97/CN/54 isolate (comprising gp120 plus the ectodomain of gp41), and monophosphoryl lipid A (MPLA), a non-toxic derivative of lipopolysaccharide that acts as a Toll-like receptor 4 (TLR4) agonist adjuvant. The vaccine aims to induce strong systemic and mucosal immune responses, including high-titer binding antibodies and balanced Th1/Th2 cellular immunity. It has been evaluated in various administration modalities, including intramuscular injection and dissolving microneedle arrays, often as part of prime-boost regimens with other HIV vaccine candidates[3][5]. The primary indication is prophylactic immunization against HIV infection.
