Overview

Co-trimoxazole + doxycycline is a combination of two antibiotics used together primarily for the treatment of certain bacterial infections where dual therapy is indicated. Co-trimoxazole (also known as trimethoprim-sulfamethoxazole or TMP-SMX) combines two agents that inhibit successive steps in the bacterial folate synthesis pathway—sulfamethoxazole inhibits dihydropteroate synthase and trimethoprim inhibits dihydrofolate reductase—resulting in synergistic bactericidal activity[2][3][4]. Doxycycline is a tetracycline-class antibiotic that inhibits protein synthesis by binding to the 30S ribosomal subunit of bacteria[9]. The combination has been studied and used particularly for diseases such as brucellosis and melioidosis, especially during eradication phases or when other regimens are contraindicated or less effective[5][8]. This regimen may be chosen based on local guidelines, resistance patterns, or patient-specific factors.
