Overview

Coagulation factor VIIa + factor X (marketed as Byclot) is a fixed-dose combination bypassing agent consisting of plasma-derived activated coagulation factor VII (FVIIa) and coagulation factor X (FX). It is primarily developed for the treatment and prevention of bleeding episodes in patients with hemophilia A or hemophilia B who have developed inhibitors against factor VIII or factor IX. The drug works through a synergistic mechanism where FVIIa and FX cooperate to enhance thrombin generation on the surface of activated platelets, effectively bypassing the need for the intrinsic pathway factors (FVIII/FIX) that are neutralized by inhibitors. Originally developed by KM Biologics (formerly Kaketsuken) in Japan, it is also being investigated for use in acquired hemophilia A, a rare autoimmune condition where patients develop autoantibodies against factor VIII.
