Overview

This is a four-drug combination regimen consisting of cobimetinib (a MEK inhibitor), dexamethasone (a synthetic glucocorticoid corticosteroid), ixazomib (an oral proteasome inhibitor), and pomalidomide (an immunomodulatory thalidomide analogue). Each drug has a distinct mechanism of action: - Cobimetinib inhibits the mitogen-activated protein kinase kinase (MEK) pathway. - Dexamethasone acts as an anti-inflammatory and immunosuppressant by binding to the glucocorticoid receptor. - Ixazomib inhibits the 20S proteasome subunit beta type-5 activity in cancer cells. - Pomalidomide modulates cereblon E3 ligase activity leading to anti-proliferative and pro-apoptotic effects in myeloma cells. This combination may be considered for relapsed or refractory multiple myeloma or other hematologic malignancies where multi-agent regimens are used[1][2][4][5][7].
