Overview

Cobimetinib + niraparib + atezolizumab is an investigational combination therapy composed of three distinct drugs, each with a unique mechanism of action for the treatment of cancer. **Cobimetinib** is a small molecule MEK inhibitor; **niraparib** is a PARP inhibitor; and **atezolizumab** is a humanized monoclonal antibody targeting PD-L1. Together, this regimen is designed to inhibit tumor cell proliferation via targeted therapy (cobimetinib, niraparib) and promote immune-mediated antitumor effects via checkpoint blockade (atezolizumab). Each component is approved for certain cancer indications individually, but the specific triple combination is investigational and studied in clinical trials for select solid tumors including melanoma[1][2][4]. Mechanistically, the combination aims to block MAPK signaling (cobimetinib), impair DNA damage repair (niraparib), and release PD-L1–mediated immune suppression (atezolizumab).
